TheM5E2antibodyreactswithCD14,an~53-55kDaGPI-anchoredtransmembraneglycoproteinexpressedathighlevelsonthesurfaceofperipheralbloodmonocytesandmacrophages,andatlowerlevelsongranulocytes.An~10-folddifferenceinexpressionlevelsbetweenmonocytes/macrophagesandgranulocytesmakesCD14ausefulMarkerfordistinguishingthesecellpopulations.CD14isalsofoundontissuemacrophages,Langerhanscellsanddendriticcells.CD14functionsasahigh-affinityreceptorforcomplexesoflipopolysaccharide(LPS)andserumLPS-bindingproteinandmodulatesLPS-dependentsignaltransductionduringtheimmuneresponsetogram-negativepathogensbyactingasaco-receptorforTLR4andMD-2.ThistriggersactivationofNF-kappa-B,cytokinesecretion,andinductionoftheinflammatoryresponse.TwosolubleformsofCD14havealsobeendescribed(~48and~55kDa).ThisantibodyclonehasbeenverifiedforpurityassessmentsofcellsisolatedwithEasySep™kits,includingEasySep™HumanMyeloidPositiveSelectionKit(Catalog#18653)andEasySep™HumanCD14PositiveSelectionKit(Catalog#18058);partialblockingmaybeobserved,aswellasEasySep™HLAWholeBloodCD33PositiveSelectionKit(Catalog#18287HLA),EasySep™HLAWholeBloodMyeloidPositiveSelectionKit(Catalog#18683HLA),andEasySep™HumanBuffyCoatCD14PositiveSelectionKit(Catalog#18088).
