Overview:
Trypsinisaserineproteasethatisproducedintheacinarexocrinecellsofthepancreas.TheenzymecleavespeptidesattheC-terminalsideoflysineandarginineaminoacidresidues.Recombinantpancreatictrypsinisawidelybiochemicaltoolusedinprocesses,whichinclude:recombinantinsulinproduction,cellculture,cellfermentation,proteinpeptidemapping,proteomicsequencingandcelldissociation.Trypsinfunctionisinhibitedbyserineproteaseinhibitors(e.g.TLCK,PMSF),andmetalchelatingagents(e.g.EDTA).Immobilizedtrypsiniscovalentlycross-linkedtoanagarosematrixcircumventingtheneedforenzymeremovalaftercleavage.Theresincanbereused10-20timesafterregeneratingwhenproperstorageconditionsarefollowed.
