Background

Thehighlyconserved,nuclear-localizedAkirin1andAkirin2proteinscriticallyregulatethetranscriptionofNF-kB-dependentgenesandarerequiredfordefenseagainstGram-negativebacteriaintheimmunedeficiencyandNF-kBpathways.Akirin1isdispensableinthemouse,andneitherknockoutmicenorcellsderivedfromthemhaveobviousdistinctivephenotypes.Incontrast,Akirin2isrequiredfordevelopmentinthemouseandknockoutofbothAkirinhomologsinmiceshowthatAkirin2isrequireddownstreamoftoll-likereceptor(TLR),TNF-aandIL-1bsignaling,andfortheproductionofIL-6.Akirin2isfunctionallyclosertothesinglegeneinDrosophila,asthehomozygousnullD.melanogasterAkirinmutantsshowasimilar,mid-to-earlyembryonicdeath.
