This NHS-ester coupling reaction is extremely easy to carry out, and proceeds in excellent yield. In a typical procedure, a gold slide (~1 cm²) covered with NHS-terminated SAM is immersed for 3 hrs in 0.5 mL of a methylene chloride (chloroform, THF, dry DMF and DMSO, absolute ethanol are also allowed) solution of a molecule bearing free amino group and 1.5 equivalents of NEt₃. When the reaction is complete, the substrate should be rinsed with pure solvent and dried under a stream of nitrogen.
